 糙皮侧耳(Pleurotus ostreatus)抗菌蛋白 Pl-09的分离纯化*

 Purification of antibacterial protein Pl-09 from Pleurotus ostreatus

摘要:  为了获得具有安全无毒的新型抗菌剂, 通过 CM?Sepharose FF、 DEAE?SepharoseFF、 QAE-SepharoseFF、 Sephacryl TMS?200High Resolution 层 析等 步 骤对 一株 食用 菌 糙皮 侧耳 ( Pleurotus
ostreatus) 液体深层培养液进行分离纯化, 得到了一相对分子量为 42 kd、 糖含量为 21  5% 具抗细菌活性的抗菌蛋白 Pl09. HPLC 分析结果表明, 该抗菌蛋白的纯度达到 99  75%, 纯化倍数为 6 350 倍. 抗菌活
性实验结果表明, 抗菌蛋白 Pl 09 对革兰阳性菌(G+ ) 的枯草杆菌、 金黄色葡萄球菌, 革兰阴性菌(G- ) 的大肠杆菌、 铜绿假单胞菌都有明显的抑菌活性. 此外, Pl 09 抗菌蛋白的最低抑菌浓度(MIC) 均明显低
于最小杀菌浓度(MBC) . 因此, 食用菌来源的抗菌蛋白 Pl09 在药品、 食品及饲料中具有广泛重要的应用价值.

Abstract:  In order to find a bacterial inhibitive drug very safe to human body or animal, an-antibacterial protein Pl-09 was purified 6 350-fold from edible mushroom Pleurotus ostreatus through a serious of chromatographic
techniques such as CM-Sepharose FF, DEAE -SepharoseFF, QAE -SepharoseFF and Sephacryl TMS-200High Resolution. The purity of Pl -09 was over 99 75% as analyzed by HPLC. The molecular weight of Pl -09 was 42kd
by SDS-PAGE, and its content of sugar components was 21 5%. The results of antibacterial analysis showed that protein Pl -09 could obviously inhibit both the growth of Gram positive bacteria such as Bacillus subtilis,
Staphylococcus aureus and that of Gram negative bacteria such as Escherichia coli and Pseudomonas aeruginosa. Among these, the results also showed that the value of MIC ( minimun inhibitory concentration) was much lower
than that of M BC(Minimum bactericidal concentrations). Our research proved that Pl?09 will have a very important application in the field of food, medicine and feeding.
