摘要:  摘要:将血管内皮生长因子C嵌合体CA65插入角蛋白14(kcratin l4, Kl4)启动子卜游,构建了嵌合体CA65转基因表达载体,通过显微注射法建立转基因小鼠.利用特异引物聚合酶链式反应法鉴定转
基因小鼠的基因型后,通过逆转录聚合酶链式反应检测嵌合体CA65的表达水平,证实成功建立了皮肤特异表达嵌合体CA65转基因小鼠.对转基因小鼠耳部皮肤进行免疫荧光染色,分析嵌合体CA65在血
管和淋巴管发育中的影响和作用.结果发现转基因小鼠的血管没有明显差异,但是嵌合体CA65可以选择性诱导淋巴管的生成,致使淋巴管增多,管径变粗.

Abstract:  Vascular endothelial growth factor(VEUF)-C/VEUF-heparin-binding domain chimeras CA65 gene was inserted downstream of keratin l4(Kl4)promoter to construct the vector which expressed at skin specially. The
transgenic mice were produced by microinjection method and the genotype was detected by polymerase chain reaction (PLR). The expression levels of the transgenic mice were detected by reverse transcription PLR. The chimeras
CA65 transgenic mice were established successfully. We whole-mount immunostained the cars of transgenic mice, and analyzed the function of CA65 on blood vessels and lymphatic vessels development and maintenance. The
chimeras CA65 was shown to induce lymphangiogenesis, and its diameter increased,but there were no statistically significant changes in blood vessels.
