摘要:  通过观察慢性丙型肝炎( CHC)患者外周血 CD4 + T 细胞和 CD8 + T 细胞表面程序性死亡受体1( PD -1) 的表达水平以及与 HCV RNA 载量的关系, 推测其临床意义. 运用流式细胞仪检测外周血
CD4 + T 细胞和 CD8 + T 细胞表面 PD ?1 的表达, 荧光定量 PCR 检测 HCV RNA 载量, 基因芯片检测HCV 基因型. 结果显示, 外周血 CD4 + T 细胞和 CD8 + T 细胞表面 PD ?1 的表达, CHC 患者较健康人明
显上调, 不同 HCV 基因型之间表达没有差异, 与 HCV RNA 载量和 ALT 无相关性. 实验结果表明持续病毒血症可能是 PD ?1 在 CD4 + T 细胞和 CD8 + T 细胞表面的高表达的重要原因, PD?1 的表达水平差异
与 HCV 复制和肝细胞的破坏程度没有必然联系.

Abstract:  We observed variation of expression of programmed death -1( PD ?1) on the surface of CD4 + T and CD8 + T lymphocytes in peripheral blood and the correlations between PD -1 and HCV RNA in chronic hepatitis C
patients, to explore the clinical significance. The PD -1 on CD4 + T and CD8 + T lymphocytes in peripheral blood were measured by flow cytometric assay, and HCV RNA was measured by real?time PCR. T he genetypes of HCV was
measured by gene chips. The expression of PD -1 on CD4 + T and CD8 + T lymphocytes in peripheral blood was higher in chronic hepatitis C patients than healthy control. T here was no significant difference in different genetypes. T here
was no correlation between the expression of PD -1 on CD4 + T or CD8 + T lymphocytes and HCV RNA or ALT. Sustained viremia of HCV may be one of important reasons of high level of the expression of PD -1 on CD4 + T and
CD8 + T lymphocytes in peripheral blood. There was no large relativity between the expression of PD -1 and replicate of HCV or the damage of hepatocytes.
